The expression of the complementary DNA encoding the human cholecystokinin B (CCK-B) receptor was studied by cloning and sequencing cDNA clones. The deduced amino acid sequence of the receptor was 318 amino acid residues long and exhibited the characteristic seven-transmembrane topology. The entire coding region of the receptor cDNA was identical to that of the cDNA coding for the rat and rabbit receptors. The binding affinity of CCK-B to the receptor was also the same as that of the CCK-A receptor with a Kd value of 1.4 nM and a Bmax of 34 pmol/mg protein. Northern blot analysis showed that the receptor mRNA was expressed in the human intestinal epithelial cell line Caco-2 and the rat pheochromocytoma cell line PC12, which have similar characteristics of intestinal and peripheral cell respectively. These results indicate that the expression of the CCK-B receptor is not restricted to the CCK-A receptor-expressing tissues and cell lines.Q:
